Resumo

The study aimed to characterize the biocompatibility and antimicrobial action of sodium alginate (SA)/tofu-basedhydrogels enriched with ozonized sunflower oil. Three formulations were developed loading the hydrogel with ozonized oil at weight ratios of 3:1, 2:1 and 1:1 (O-3-1, O-3-2 and O-3-3 SA/tofu, respectively). The physicochemical characterization of the hydrogels was done by Fourier-transform infrared spectroscopy (FTIR). Antimicrobial susceptibility testing (AST), minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) were used to determine their antimicrobial potential against skin bacteria (Staphylococcus aureus, S. epidermidis, Streptococcus pyogenes and Escherichia coli). The biocompatibility of the formulations with fibroblast (GM07492-A) was characterized by XTT and wound healing assays. The identification of a peak at 1000 cm-1 in the spectrum of the ozonized hydrogels confirmed the successful oil-loading. Non-ozonized hydrogel did not show antimicrobial action (MIC and MBC > 100%), while the three ozonized formulations inactivated the four bacteria (MBC and MIC ranged from 0.39 to 50.0%). The viability of fibroblast cells remained unchanged (p > 0.1) after treatment with O-3-1, O-3-2 or O-3-3 SA/tofu. Similar to non-ozonized formulation (p < 0.001), complete wound closure was detected 48 h after treatment with SA/tofu O-3-1. The ozonized hydrogels were biocompatible with fibroblasts and showed germicidal action against skin bacteria. (AU)
