Resumo

In this study, we investigated the influence of limonene concentration (1-5 %, w/w) and type of oil phase (isopropyl myristate or tricaprylin) on the physicochemical and irritative properties of nanoemulsions (NEs), as well as their ability to improve the skin penetration and cytotoxicity of the retinoid fenretinide. Our goal was to contribute to the development of nanoformulations for topical administration to the breast skin and prevention of breast cancer recurrence. The NE exhibited diameter below 200 nm (PDI <0.2), negative zeta potential (-9 to -11 mV), and did not cause vascular damage in the choriollantoic membrane of eggs regardless of limonene concentration and oil phase type. However, only NEs with isopropyl myristate as the oil phase and 1-2.5 % limonene remained stable for 6 months, and incorporation protected limonene from evaporation. Fenretinide skin penetration increased with time and limonene concentration; NEs containing 2.5 % limonene provided the highest (5.8-fold) fenretinide penetration into viable skin layers at 12 h. Incorporation in the limonene-containing NEs increased fenretinide cytotoxicity against MCF-7 cells, and reduced the migration of MCF-7 and MDA-MB-231 cells (1.5-2-fold) at non-cytotoxic concentrations, supporting the NE potential applicability for topical preventive and adjuvant therapy of breast cancer.
