Hydrogen production through dark fermentation represents a promising strategy for sustainable energy recovery and the valorization of agro-industrial wastewater streams. This study investigates the use of soybean molasses - by-product of soybean processing - as a substrate in thermophilic anaerobic bioreactors operated at 50, 55, and 60 degrees C, aiming for the co-production of hydrogen and carboxylic acids. The reactor operated at 50 degrees C with a high organic loading rate (26 kg-COD/m3/d) achieved the highest process efficiency, with 75 % carbohydrate removal, a hydrogen productivity of 51.4 mol-H2/m3/d, and a yield of 4.6 mol-H2/kg-carbohydrate. Increasing the temperature to 55 and 60 degrees C led to pronounced reductions in both hydrogen productivity (by 32 % and 68 %) and carbohydrate removal (by 51 % and 57 %, respectively). The microbial community analysis based on 16S rRNA gene PCR revealed temperature-driven shifts in composition, explaining the observed changes in process performance. Optimal hydrogen production at 50 degrees C was associated with a diverse microbial community, including hydrogen-producing genera such as Thermoanaerobacterium, Clostridium sensu stricto 11, and Ethanoligenens. These findings demonstrate the feasibility of using soybean molasses as a substrate in high-rate thermophilic fermentation and highlight the critical role of microbial community structure in process performance, reinforcing the potential of this approach for integrated bioenergy and biomolecules production.
