Aspergillus flavus is a major concern in cereal production due to its ability to produce aflatoxins, highly carcinogenic mycotoxins. This study assessed the antifungal and anti-mycotoxigenic activity of oregano (OEO), thyme (TEO), and for the first time, a novel Tween 80-based emulsion combining thyme and oregano essential oils (OTEO) against A. flavus and in maize kernels stored for 30 days. Chemical characterization revealed monoterpene-rich profiles: OEO was dominated by carvacrol (64.7 %), TEO by thymol (45 %), while OTEO presented an intermediate composition (carvacrol 32.9 %, p-cymene 25 %, thymol 14.3 %). OEO and TEO showed identical MIC values (0.7 mg/mL), with TEO exhibiting greater fungicidal activity (MFC 1.3 mg/mL) than OEO (2.8 mg/mL) in in vitro assessment. OTEO displayed lower activity in non-emulsified essential oil form, but at 2 x MIC, and all oils caused similar to 98 % cell death and ergosterol depletion. Emulsification enhanced efficacy, yielding stable formulations (179-210 nm, zeta-potential < -40 mV) with significantly reduced MIC/MFC values (0.15-0.6 mg/mL). In maize assays, non-emulsified essential oil partially reduced fungal growth and aflatoxin levels but failed to prevent contamination. In contrast, the novel OTEO emulsion, together with the OEO emulsion, completely suppressed A. flavus growth and AFB1 + AFB2 production throughout 30 days of storage. The TEO emulsion also reduced contamination, keeping aflatoxin levels at 2.6 mu g/kg, below regulatory limits. These findings highlight the innovative potential of essential oil emulsions-particularly the novel thyme-oregano combination-as effective natural alternatives to control A. flavus and aflatoxin contamination in stored maize.
