This study reports the formation of a novel salt and solvate of mefenamic acid (MFA) with tromethamine (TRIS) with improved solubility. Mefenamic acid is a fenamate used as an anti-inflammatory in the treatment of muscle pain, arthritis, and dysmenorrhea. Due to its low aqueous solubility, multicomponent solid forms are an alternative to optimize its physicochemical properties without altering its therapeutic effect. Through the mechanochemical method, two new multicomponent solid forms were obtained, compounds I and II, formed by stoichiometric portions of MFA and TRIS assisted by minimal amounts of ethyl acetate and acetonitrile, respectively. Single crystals of compounds I and II were obtained through slow solvent evaporation and allowed their crystalline structure to be determined, revealing a pharmaceutical salt (compound I) and an ionically stabilized solvate (compound II). The background of the compounds was analyzed through, powder X-ray diffraction, infrared absorption spectroscopy, and simultaneous thermogravimetry-differential thermal analysis and differential scanning calorimetry analyses. Solubility studies were performed using the shake-flask method, and the compounds were quantified using UV-Vis spectroscopy with a validated methodology, revealing an increase of 2.725 (compound I) and 2.390 (compound II) compared to isolated MFA. Thus, the obtaining and characterization of two new multicomponent solid forms of mefenamic acid demonstrate the relevance of crystal engineering in pharmaceuticals, particularly regarding the influence of the crystal lattice and solvent on their physicochemical properties.
