Resumo

The objective of this work was to transpose the synthesis of the drug Pioglitazone, used in the treatment of type 2 diabetes mellitus, from the conventional batch process to flow synthesis in capillary microreactors. This included defining the number of parallel microreactors (nMR), which indicates how many microreactors are necessary to achieve the same production as the batch process, serving as a guide for the industrial application of microreactor technology. Additionally, we aimed to determine the kinetic and thermodynamic parameters of each reaction involved in forming intermediate products. The synthesis route was also improved by reducing one step in the complete path. We successfully transposed the synthesis of intermediate products 2 and 4 to the flow process, achieving 66 % and 85 % yields, respectively. In contrast, the new reaction for forming intermediate 3 in batch yielded 54.2 %. For intermediates 2 and 4, the optimal nMR values obtained were 4.4 and 2, respectively. We synthesized the drug Pioglitazone using a homemade batch reactor for hydrogenation, and HPLC-UV analysis indicated that the purity of the synthesized Pioglitazone (91.5 %) is close to that of the commercial Pioglitazone (95.5 %) used as a reference. These results suggest the potential for scaling up the manufacture of this drug with the assistance of microreactor technology. (AU)
