Abstract

The pathways and biological processes associated with embryo competence, cryosurvival and the success of obtaining an embryo-derived pregnancy are still lacking. The present work aims to elucidate strategies and understand the mechanisms of the competence of in vitro produced (IVP) bovine embryos in a holistic approach. In a series of three experiments, we will look for: 1) identify differentially expressed molecules between inner cell mass (ICM) and trophectoderm (TE) cells from embryos that resulted in positive and negative pregnancies for the establishment of the transcriptional profiles and pathways that favor embryo-derived pregnancy success according to the cell type; 2) understand the functional role of facilitating pregnancy molecules according to cell type (ICM and TE) during early embryo development; 3) Establish an embryo culture medium that favors the activation of facilitating pregnancy molecules and pathways in order to improve embryo in vitro production system. Multivariate and univariate statistical models will be used in robust softwares capable of analyse a great amount of data and provide solid answer to the tested hypothesis. The significance level of 5% (P<0.05) will be used. At the end of the project, a comprehensive and reflexive analysis of the results will be done to establish the molecular pathways that are determinant for embryo competence, as well as the establishment of strategies to increase the efficiency of in vitro production in order to achieve the success of embryo-derived pregnancy, favor the dissemination of IVP embryos in the cattle reproductive management, and the production of animal protein.
