Abstract

The electronic cigarette (e-cig) was developed to promote and facilitate smoking cessation, however, the relationship of risks and benefits as well as the effects of e-cig on the oral mucosa are unknown. The objective of this study is to evaluate the effect of using e-cig on saliva and the oral mucosa of its users. Two groups will be composed: Electronic Cigarette Group (GCE) - 48 regular and exclusive e-cig users; Control group (GCO) - 48 non-smoking and non-e-cig users, matched in gender and age to the GCE. All participants will be examined. Afterwards, unstimulated saliva will be collected for the evaluation of the salivary metabolome. In addition, 2 ml of the liquid present in each participant's e-cig will be collected for the evaluation of their composites. Both analyzes will be done by means of gas chromatography coupled to the mass spectrometer (GC-MS). Saliva will also be submitted to vibrational spectroscopy analysis (Raman dispersive, FT-Raman, FTIR). The cytological smear will be done on the left side of the tongue for citochemical evaluation and, on the right side, for gene expression evaluation. The exfoliative cytology will be evaluated by the Papanicolaou method (cytomorphological evaluation - inflammation and cellular maturation), Silver staining (evaluation of nucleolar organizer regions - AgNORs) and by qRT-PCR (CYP1A1, CYP1A1, CYP2A6, CYP2E1, L1B, IL6, CXCL8, TNF, CCL2, PTGS2, KRT1, KRT5, KRT13, KRT15, TP16, TP21 and TP53). The data will be treated by Student t-test (parametric data) or Mann-Whitney test (non-parametric data). For metabolic analysis, will be performed in addition, the principal component analysis (PCA) and ROC curve. The associations between variables and clinical data will be explored using Spearman's correlation coefficient. For all analyzes, a significance level of 5% will be adopted. (AU)
