Abstract

The periodontium is a group of structurally complex tissues composed of several cell types with varied functions. In periodontal disease there is a progressive process of tissue loss, resulting in irreversible aftereffects, which can culminate in tooth loss. Tissue engineering mediated by stem cells may be a therapeutic alternative for future, aiming at regenerating the lost tissues. Stem cells from the periodontal ligament have been extensively studied, but there is still no consensus on their properties and definitions. It is already well established that periodontal ligament stem cells correspond to a subgroup of cells within a heterogeneous cell population. Furthermore, enriched populations of these cells have better regenerative and therapeutic properties. Thus, it is necessary to establish protocols for obtaining, isolating and expanding cell cultures, ensuring more homogeneous and reproducible results. We propose the use of injectable platelets rich-fibrin (i-PRF) as a mobilization factor to enrich, in vitro, population of cells isolated from the periodontal ligament. The subpopulations will be characterized in vitro in terms of immunophenotyping, multidifferentiation capacity, proliferation and senescence. Afterwards, we will establish cell sheets that we expect to be more efficient and evaluate their potential to regenerate the periodontium in furcation defects in Wistar rats. (AU)
