Abstract

Brain ischemia is one of the most relevant of the human diseases, with high mortality and morbidity rates worldwide. It is a frequent condition in clinical practice that has no simple therapeutical solution, mainly due to the difficulties in performing a physiopathological study, which arise from the great diversity in its anatomical localization, its etiology and clinical manifestations. The aim of this work is to evaluate the expression pattern of genes related to apoptosis (NMDA, FAS, c-FLIP, caspase-8, caspase-3 and HSP70), either alone or associated to the neuroprotective effect of hypothermia and/or ketoprofen. To achieve this goal we will use an experimental model in which rats will be induced to develop focal brain ischemia by the occlusion of the midlle cerebral artery with an intraluminal probe and the results will be assessed by semi-quantitative polymerase chain reaction (RT-PCR) and real-time quantitative polymerase chain reaction (RQ-PCR). Wistar adult rats will be divided in three experimental groups: 1) control group, which comprises animals that will receive anesthesia only; 2) sham group, which comprises animals that will undergo a mock surgical procedure; 3) ischemic group, which will be further divided into four subgroups: Ischemic (SgI), Ischemic with hypothermia (SgHI), ischemic with ketoprofen (SgIC), and ischemic with hypothermia and ketoprofen (SgIHC). All animals within the ischemic subgroups will be induced to develop brain ischemia for one hour, sacrificed, and samples will be collected and further analyzed. (AU)
