Influence of the type of abrasive and fluoridated salt on the metabolism and uptake of fluoride in young rats

Abstract

The aim of the present study is to evaluate fluoride metabolism and uptake in young rats, after administration of fluoride as dentifrices containing different fluoride salts (NaF or MFP) and abrasive systems (silica or calcium carbonate). For this purpose, 80 5-day-old rats will be divided into 4 groups (n=20/group). The difference among the groups will be the type of dentifrice solution (slurry of 1 g dentifrice : 3 mL deionized water) to be administered to the animals by gastrogavage from days 5 to 10, as follows: group 1 (negative control): placebo dentifrice (without fluoride or abrasive); group 2: dentifrice containing 1,500 ppm fluoride as MFP and calcium carbonate (45%) as abrasive; group 3: dentifrice containing 1,500 ppm fluoride as NaF and silica as abrasive; group 4: dentifrice containing 1,500 ppm fluoride as MFP and silica as abrasive. Each day during the experimental period, the animals will receive the dentifrice solutions by gastrogavage, in a dose corresponding to 1,0 mg fluoride per Kg body weight. After the last gavage (day 10), the animals will be transferred to metabolic cages for 12 hours in order to allow collection of urine and feces for fluoride analysis. Then the animals will be euthanized and plasma and femur will be collected for fluoride analysis. The fluoride analyses will be done by the direct method or after hexamethyldisiloxane-facilitated diffusion. Data will be checked for normality and homogeneity for the selection of the appropriate statistical test (p<0.05). (AU)
