Identification of genetic and epigenetic alterations in adenoid cystic carcinoma with high-grade transformation

Abstract

Recent research indicates that the silencing of tumor suppressor genes by DNA methylation may be involved in tumor progression. In literature, there are few molecular studies about the transformation into high-grade of adenoid cystic carcinoma and none of them reports the possible involvement of DNA methylation. Adenoid cystic carcinoma is one of the most common salivary carcinomas and this transformation is related to a more aggressive clinical course and increased incidence of lymph node metastases. The aim of our study is to investigate in adenoid cystic carcinoma with high-grade transformation the genetic alterations and DNA methylation using the technique of MLPA (Multiplex Ligation Probe Amplification). This analysis may contribute to understanding the pathogenesis of this phenomenon and to verify the possible existence of a marker gene that would help in identifying of the disease. Moreover, DNA methylation is an attractive target for the development of new therapeutic approaches, because of gene silencing by epigenetic alteration is potentially reversible. (AU)
