Abstract

Cutaneous mast cell tumors (MCTs) commonly occur in dogs and account for 7 to 21% of all canine cutaneous neoplasms. These tumors range from a benign mass that can be cured by surgical excision alone, to potentially fatal metastatic disease. The histologic grading system proposed by Patnaik et al. (1984) is currently used to classify MCTs and it is based on certain histomorpholologic features. Although this grading system is the accepted method for predicting MCT behavior, it is not reproducible, with as much as 50-60% discordance between experienced pathologists. A novel two-tiered grading system was recently proposed which separated high from low grade tumors (Kiupel et al. 2011). Using this new system, there were significant associations between grade and time to metastasis and survival time. As the sequence of the dog genome is more similar to that in humans than rodents. This study aims to verify the genomic copy number alterations in MCTs using the technique CGHarray . Candidate genes will be validated by immunohistochemistry. Frozen samples of eight high grade and eight low grade MCT will be used in the study to extract DNA topeform the technique CGH array. (AU)
