Abstract

The correct development of the gastrointestinal tract depends on proper suckling-weaning transition. In the gastric mucosa, hormones and milk-born peptides regulate cell proliferation, differentiation and death. Our hypothesis is that suckling is one of the main regulatory agents during development, and so, changes in dietary pattern or conditions can alter the action of molecules that control growth. In order to test this idea, this proposal aims to evaluate the effects of fasting and early weaning on the expression, distribution and activity of corticosterone, ghrelin and TGFbeta in the gastric epithelium to ultimately investigate the integration of these elements. On the 3rd week of postnatal development, rats will be submitted to fasting or early weaning and the stomach will be collected at different ages to extract RNA, protein and tissue for RT-PCR, qPCR, microarray, protein multiplex array, immunoblot and immunohistochemistry to detect: the expression of genes and proteins; cell signaling; cell proliferation and differentiation and cell cycle control. Results will be put together according to experiments and statistically analyzed. (AU)
