Characterization of SXT element of bacterial isolates obtained from environmental samples

Abstract

Antimicrobial resistance has become a worldwide problem. The mechanisms of antibiotic resistance in bacteria are mediated primarily through mobile genetic elements that carry resistance genes. The SXT element, first isolated in an EGM V. cholerae, has contributed to the spread of resistance genes in different bacterial genus. The first reported observation of the SXT element in South America was held at the Laboratory of Public Health of USP, where these elements were observed in two strains of V. fluvialis. Objective. Characterize and map the SXT elements previously observed in V. fluvialis and expand the investigation of the presence of such elements in isolates from different sources obtained from 1994 and belonging to the collection of cultures of the Public Health Laboratory -FSP-USP. Methods. The existing SXT elements in V. fluvialis (isolates FSP286/08 and FSP561/08) will be characterized using the complete genome sequencing. Investigation of the presence of these elements will be carried out in about 800 isolates belonging to a culture collection of the Laboratory. Positive strains will have antibiotics susceptibility profile determined, and presence of resistance genes investigated. Expected results. Comparing the Brazilian variant SXT element with those existing in epidemic strains available in GenBank, and to design specific primers for Brazilian SXT variants from the analysis of these sequences.
