Evaluation of seminal plasma proteins and its correlation with fertility of stallions

Abstract

The equine species has reduced fertility rates when compared to other domestic species. Part of these observations relates to the fact that horses are not selected for fertility. In addition, there are tendencies to attribute infertility problems to the female, which compromise a detailed evaluation of the male. Among the factors that can alter the reproductive indices, sperm quality takes a considerable effect. It has been shown that some components of seminal plasma are related to the fertility of bovine semen, suggesting that the determination of these components could be considered a form of evaluation of the ejaculate. Thus, sperm kinetic parameters, plasma membrane integrity and acrosomal, lipid peroxidation, DNA integrity, spermatic capacitation, absolute plasmatic membrane potential measurement and the verification and quantification of the proteins osteopontin and semenogelin in sperm from fertile and subfertile stallions will be compared. Moreover, in order to identify proteins in the seminal plasma of fertile and subfertile stallions that are supposed to be correlated with fertility, two-dimensional electrophoresis and mass spectrometry will be performed. Isolation and characterization of proteins present in seminal plasma may permit evaluation of its influence on sperm cells and to identify safe and early fertile stallions in relation to those subfertile. And likewise, enable the development of new techniques that may contribute to the increase and prediction of fertility in stallions. (AU)
