Abstract
Primordial germ cells (PGC) are the precursors of the gametes. In mice, PGC emerge from a pool of epiblastic cells around 7dpc. During migration from the epiblast to the gonads, the PGC undergo epigenetic reprogramming that control the expression of PGC genes. This reprogramming includes the silencing of retrotransposons. This silencing is important to maintain the integrity of DNA. The mouse is the model of choice for the studies on PGC because of the similarity between the human and the mouse genomes. However, it studies have been suggesting that the rat is more similar to humans than the mouse. In recent study our group showed that the expression of MVH, which is specific for germ cells, in the rat is similar to what was described for human PGC and different from mice. In mice the expression of MVH begins after gonad colonization and seems to be related with the expression of retrotransposons and of PIWI proteins. In the rat we observed that MVH is expressed before gonad colonization by the PGC. Thus, the aim of this study is to investigate the relationship between MVH , piRNAs and retrotransposons expression in the rat PGC. This study could contribute to the elucidation of the mechanisms that control rat germ cell development and could be compared to what is observed in mice and humans. (AU)
