Endothelial evaluation of contraceptive formulations by hemostatics, vasoactive and inflammatory parameters

Abstract

More than 100 million women worldwide make use of hormonal contraceptives. Among them, we point combined oral contraceptives (COC)formed by estrogens and progestins. Despite its desired effect on contraception, COC have their metabolism in the liver, involving the synthesis of coagulation proteins as well as modulating the expression of adhesion molecules, and vasoactive factors present in the endothelium predisposing to venous thromboembolism among users. To combat these side effects, different formulations of different estrogens and progestins have been researched. Objective: To evaluate the effect of two contraceptive formulations consist of: 1 - Levonorgestrel (150 mcg) combined with Ethinyl estradiol (30 mcg) and 2 - Estradiol Valerate (1,2 or 3 mg) in combination with Dienogest (2 or 3 mg), in cultured endothelial cells HUVECs and HAECs. Materials and Methods: After stimulation of these cells by the compounds cited, we will determine the cell supernatant and homogenized in the following parameters: NO, Endothelin E, Prostaglandin, Tissue Factor, VCAM1, ICAM1 and E-selectin by different methods (ELISA immunoassay, flow cytometry, MTT). Thus, we evaluate the different contraceptive formulations vasoactive factors, inflammatory and haemostatic by culture of endotelial cells. (AU)
