Abstract

The aim of this animal study will be to evaluate by comparative analysis of the biochemically, tomographic, photometrically, histomorphometrically and immunohistochemically study, the influence of chemotherapeutic drugs on the evolution of experimental periodontitis (EP) in rats. Following a table generated by a computer program, 180 male rats will be divided into 6 groups, with 30 animals each. SPE- SS group, also called the sham group. The animals in this group will receive two intraperitoneal injections with 48-hour interval between them of 0.5 ml of saline solution 0.9 % (SS). These animals will not be induced to EP. EP - SS group: the animals in this group will receive two intraperitoneal injections with 48-hour interval between them of 0.5 ml of SS. Soon after the first injection, PE will be induced in the lower first right and left molars. SEP - 5FU group: the animals in this group will receive two intraperitoneal injections with 48-hour interval between them of 60 mg / kg and 40 mg / kg of 5-Fluorouracil (5-FU) respectively. These animals will not be induced to EP. EP - 5FU group: animals in this group will receive two intraperitoneal injections with 48-hour interval between them of 60 mg/kg and 40 mg/kg 5-FU respectively. Soon after the first injection, PE will be induced in the lower first right and left molars. SEP - CIS group: the animals in this group will receive two intraperitoneal injections with 48-hour interval between them of 5 mg/kg and 2.5 mg/kg of cisplatin (CIS) respectively. These animals will not be induced to EP. EP - CIS group: the animals in this group will receive two intraperitoneal injections with 48-hour interval between them of 5 mg/kg and 2.5 mg/kg CIS respectively. Soon after the first injection, PE will be induced in the lower first right and left molars. EP was induced by the placement of a number 24 cotton thread around the lower first molars. After of 07, 15 and 30 days after the first intraperitoneal injection (SS or chemotherapy) animals will be euthanized using an overdose of thiopental (150mg/kg). Blood will be collected via cardiac puncture for hematology and biochemical analyzes, at the following times: prior to application of chemotherapy or SS and euthanasia periods (7, 15 and 30 days). Blood samples will be divided into two microtubes, one of them for the blood test and the other for the biochemical analysis of AST, ALT, creatinine and urea. The mandibles collected will be divided into two parts and processed according each analyzes. For the micro-computed tomography (Micro-CT) and photometric analysis will be assessed the loss of alveolar bone volume. The contralateral mandibles are used for histomorphometric analysis which will be evaluated and the percentage of bone histology in the furcation area, and immunohistochemical analyzes of biomarkers in the furcation area (RANKL, OPG, TRAP, BAX, PCNA, HIF1±). Quantitative data will be analyzed statistically (p < 0.05) in a specialized computer program (Bioestat 5.0). (AU)
