Dental pulp cell differentiation and biomineralization: cell culture assays, experimental models and clinical application in human

Abstract

The advances in the development of materials for conservative pulp tissue therapy has allowed the preservation of the vitality of tissue and tissue healing through the mineralized neoformation. Biodentine" and MTA are dental materials used for this purpose, although the mechanisms invoved in biomineralization induced by these materials are not fully understood. Therefore, further studies should be conducted both in vitro as in vivo to shed light on this important biological repairing process through investigation of the molecules and signalling pathways involved. For this purpose, to dissect possible mechanisms of inflammation and repair, in vitro models of human dental pulp cell culture will be used and in vivo biompatibility will be investigated in the subcutaneous tissue of mice and in dogs teeth. In a second phase, we aim to evaluate, in a randomized clinical study, if the materials showing histological satisfactory results could be indicated for pulpotomy in humans primary teeth. Finally, in an animal model, we will evaluate whether the deposition of mineralized tissue induced by dental pulp cells could be mimicked by periodontal ligament cells to stimulate the repair after furcation perforation, which still represents a clinical situation difficult to be treated.
