Abstract

With the limitations of the use of grafts or autogenous bone transplantation, biomaterials have been studied to replace them in the clinical application as implants. In Tissue Engineering, the use of a cell source is coupled with the construction of three-dimensional frameworks as a clinical alternative that can be applied to patients with bone loss problems.This study aims to evaluate mesenchymal stem cell (MSCs) in in vitro biocompatibility tests with polymeric materials developed as scaffolds for bone tissue bioengineering.We will use as biological control Vero cells, an established cell line recommended internationally for studies with biomaterials. The scaffolds will be made of bioresorbable polymers composed of poly (hydroxybutyrate-co-hydroxyvalerate) [PHVB] and poly (e - caprolactone) [PCL] blends in ratio of (75/25) and (50/50).We will use the pure polymers as efficiency control to their blends. The materials will be characterized by scanning electron microscopy (SEM), stereoscopy, micrometer and FTIR. The biological evaluation will be done through the analysis of MSCs and Vero cells, both cultured on different scaffolds, evaluated by light microscopy, where will be made morphological and cytochemical analysis, and scanning electron microscopy. We also will perform biochemical assays for measuring the relative amount of adherent viable cells (MSCs and Vero cells) by the MTT method as well as the activity of alkaline phosphatase enzyme produced by MSCs. Finally, we will make the culture medium analysis of MSCs. (AU)
