Abstract

The immunotherapy has been considered important therapeutic tool in the treatment of malignancies. The antiangiogenic strategy is among the most promising approaches. The vascular endothelial growth factor (VEGF) plays a critical role in the angiogenic process mainly in solid tumors. Bevacizumab is an anti-VEGF humanized monoclonal antibody, which has been used in treating a variety of tumors. However, adverse side effects that may impair their use were observed. Experiments conducted by our group showed promising results when the anti-Bevacizumab idiotype mAb (Id) 10.D7, obtained in our laboratory (Process FAPESP 2009 / 18631-1), was used as immunogen. This project proposes to expand the study, which has shown, in proof of concept experiments, survival increase of annimals that received tumor implantation after being immunized with anti-Id mAb 10.D7. Thus, we intend to analyze the ability of the mAb 10.D7 to inhibit metastatic tumor proliferation, and to construct the single chain antibody (ScFv) 10.D7 from the isolation of the variable region genes of the light and heavy chains of 10.D7 hybridoma to evaluate its potential as a genetic vaccine.
