Spinal cord injury and male infertility: effect of probenecid treatment on seminal toxicity of prostatic origin.

Abstract

Men with spinal cord injury face many difficulties when trying to have children. Besides erectile and ejaculatory dysfunction, which can be effectively treated, these men also present low semen quality. In order to better understand the mechanisms of which these men present low semen quality, it was verified that the seminal plasma present toxicity caused by inflammasome complex. This complex can be blocked by oral administration of probenecid, increasing progressive motility in these men. After seminal plasma proteomic studies of oral probenecid treated men, realized in our laboratory, it was possible to suggest a panel of proteins that can be responsible for increased sperm motility after treatment. In order to continue to understand the mechanisms affecting low semen quality, the aim of the present study is to investigate the effect of treatment with oral probenecid on seminal toxicity of prostatic origin. For that, 15 spinal cord injury men will be treated with 1000 mg in total by day of oral probenecid. One sample will be collected, by penile vibratory stimulation, before the beginning of treatment and another sample by the end of treatment. One aliquot will be used for semen analysis and the other amount will be used at protein analysis. For protein validation ELISA and Western blotting techniques will be performed. For group comparison paired (Pre x Post) and unpaires (Pre x Control) Student t test will be performed (p < 0.05). With these results we hope to better understand the mechanisms of which the seminal plasma of SCI men is toxic for the sperm. (AU)
