Abstract

Today most complex biopharmaceuticals manufactured for human therapies are made commercially by use of the Chinese Hamster Ovary (CHO) cells as the "factory". Whilst the CHO cell line performs many of the required modifications (PTMs) during synthesis, specific PTMs (required for many proteins) are not performed. Human cells, with the capacity to perform human-specific PTMs, will present much more reassured breadth of PTM for target biopharmaceuticals in the emerging product pipeline. There are a number of human cell lines that have better capability to generate appropriate PTM but these need to be fully characterized and require maximization of biomass attainment. This is the focus for the proposal that links together the research backgrounds of Profs Swiech and Dickson. In brief the objective of this proposal is provide support to investigate how to develop a portfolio of collaborative grant applications that will (3) investigate the profile of PTMs performed by a series of human-derived cell lines, and(4) outline bioprocess development to enable scale-up and harvest of specific biopharmaceuticals from specific cell lines at commercially-viable biomass.
