Regulation of dentin-pulp and periodontal ligament cells in HELTHY individuals and with hypophosphatasia

Abstract

The aims of this study will be: (1) To determine basal differences in gene expression in pulp vs. periodontal ligament (PDL) cells in healthy and hypophosphatasia-diagnosed (HPP) subjects, focusing on genes associated with differentiation/mineralization and inorganic phosphate (Pi) metabolism; (2) To determine the effect that Pi treatment exerts on pulp vs. PDL cells, in vitro. To obtain primary cells cultures of the pulp and PDL, patients between 16 and 20 years old that have third molar submerged extracted for orthodontic reasons will be identified and classified at two groups: A - control group (healthy subjects), B - test group (hypophosphatasia-diagnosed subjects). Proliferation/viability assay and mineralization in vitro, and evaluation of mRNA with a quantitative real-time PCR will be performed. Mean and standard deviation (S.D.) will be calculated to establish statistical comparisons at a level of significance of 5%. (AU)
