Abstract

Hypertension is a common cardiovascular disease that affects millions of people worldwide. There are several manners to treat hypertension, such as the use of angiotensin I converting enzyme (ACE) inhibitors. In the last years, it has been demonstrated that some plant species can inhibit the ACE and the rosmarinic acid is one of the compounds of these species. Then, the aims of this study are to verify if the rosmarinic acid presents inhibitory effect on ACE activity, and to compare the rosmarinic acid effect with that of captopril in normal and hypertensive rats. Thus, the animals will be randomly divided into two groups: hypertensive (n = 21) and normotensive (n = 21). The hypertensive rats will undergo a surgical procedure (2 kidneys - 1 clip) for hypertension induction, while the normotensive animals will be sham-operated. Three weeks after these surgeries, the possible inhibitory effect of rosmarinic acid on the ACE will be tested and compared with the captopril effect. To assess the in vivo ACE activity, dose-response curves for angiotensin I and II will be done simultaneously to blood pressure measurements. The in vitro ACE activity will be measured in plasma, fluorimetrically, using the substrate hipuril-histidil-leucine.
