Influence of the type of abrasive and fluoridated salt on the metabolism and uptake of fluoride in young rats

Abstract

The aim of this study is to evaluate the metabolism and incorporation of fluoride in young rats after administration of fluoride in toothpaste form containing different fluoride salts (NaF or MFP) systems and abrasive (silica or calcium carbonate). For this purpose, 80 rats with 5 days of life will be divided into four groups 20 animals each. The difference between the groups is the type of solution toothpaste to be administered to animals by gastric gavage from 5th to 10th day life, namely: - Group 1 (negative control): placebo dentifrice (without fluoride or abrasive), group 2 dentifrice containing 1,500 ppm fluoride in the form of MFP and calcium carbonate (45%) as an abrasive, group 3: toothpaste containing 1,500 ppm NaF as fluoride and silica abrasive, group 4: toothpaste containing 1,500 ppm fluoride in the form of MFP and silica abrasive. Every day, animals receive solutions dentifrice gastric gavage at a dose corresponding to 1.0 mg of fluoride per kilogram of body weight. After the last gavage (10 days), animals will be transferred to metabolic cages for 12 hours to collection of feces and urine for analysis of fluoride. Then they will be euthanized, being collected plasma and femurs for analysis of fluoride. Analyses of fluoride will be performed by the direct method or after facilitated diffusion hexamethyldisiloxane. The data will be checked for normality and homogeneity, for selecting the appropriate statistical test (p<0.05). (AU)
