Abstract

The project aims the development of metallomic studies related to breast milk in different stages of lactation. To accomplish that, mapping and quantification of calcium, iron and zinc - minerals of great importance to newborn's health and nutrition - will be made at breast milk proteins, as well as their characterization. The milk, collected from healthy women from Botucatu-SP, will be submitted to two-dimensional electrophoretic procedures with polyacrylamide gel (2D PAGE) to fractionation and identification of proteins that bound calcium, iron and zinc, as well as analytical procedures - qualitative and quantitative - to evaluate metal ions in the protein spots. The proteins spots in which the metal ions of interest are detected/quantified will be characterized by MALDI-TOF MS after tryptic digestions of these spots. It's expected, through a series of techniques never used before for this kind of sample, analyze the distribution of calcium, iron and zinc in breast milk proteins inserted in an environment where they are subjected to the influence of other proteins and biomolecules, verifying, besides other aspects, to what proteins they bound preferably and in which quantity. It's also expected to contribute at the elucidation of the component's physiological and functional aspects. (AU)
