Influence of irrigants gel and the ultrason at the biofilm elimination

Abstract

The microbial biofilms and their products are the main risk factors of onset, propagation and perpetuation of chronic apical periodontitis. The application of chemical disinfectant solutions, among them, chlorhexidine and sodium hypochlorite in different concentrations, are conventionally used for antisepsis of the root canal system, but there are no reports of sodium hypochlorite in the form of gel, when applied for this purpose. In addition to ultrasonic irrigation has been suggested to disinfect areas inaccessible to endodontic instruments. Thus the objective of this research will evaluate the antimicrobial action of sodium hypochlorite gel 1% and 2.5% chlorhexidine gel 2% and 24% EDTA gel, ultrassonoenergizados or not, when kept for 5 minutes on the biofilm Oral. For the study will be used bovine dentin blocks sterilized, placed in a dental appliance that will be used by a volunteer for 3 days for the formation of oral biofilm. After that the blocks are washed with 100 mL of PBS Ecora with 50 microliter of a solution of Syto 8/Iodeto propidium (Live / In Vitro Deadtechnique-Gen). After staining, the samples will be evaluated in Confocal Microscope Laser Scanning. The images obtained are analyzed by specific software to evaluate the thickness, the thickness distribution, the percentage of colonized area in each section and biofilm biomass. After this preliminary analysis, the dentin blocks colonized by biofilm ¼ldos irrigators will receive 100 experimental and control solution, keeping them in contact for 5 minutes. In groups where they will receive ultrassonoenergization, this will be held for 1 minute. After the application of irrigation, the specimens will be re-examined microscopically, evaluating the parameters mentioned above. The pre-and post-irrigation obtained will be statistically analyzed, which will suggest which solution is more action on the oral biofilm, as well as a potentiation of the same will occur with the use of ultrasound.
