Abstract

5-fluorouracil (5-FU) is an analogue of uracil antimetabolite widely used to treat head and neck cancer. The cytotoxic mechanism is well established in the literature, but little is known about the effects the drug has on cellular proteins and metabolites. The objective of this study is to analyze the tumor-secretome profile treated with 5-FU and evaluate their effects on cells of the same origin. As strategy, SCC9 line (tongue's carcinoma) will be treated with the drug 5-FU and have your conditioned medium collected and analyzed by mass spectrometry and magnetic resonance for proteins and metabolites, respectively. This conditioned medium is used in experiments to analyze the secretoma's effects stimulated by the drug 5-FU. Three genes related to signaling pathways of the products with differential expression will be selected and will have their expression validated by real-time PCR.
