Use of antioxidants in Botu-crio® diluent for freezing sperm cells from epididymis of cats undergoing elective orchiectomy

Abstract

The use of domestic cats is an excellent choice as an experimental model. The possibility of sperm cryopreservation bring large benefits, especially for the conservation of wildlife. The purpose of this study is to assess the suitability of the use of a national diluent (Botu-crio®) for the cryopreservation of cat sperm. Moreover, to evaluate if the addition of vitamin E at different concentrations to the botucrio diluent is more effective for cryopreservation of semen obtained from epididymal of domestic cats. Fifty four tom cats will be used in the experiment after being submitted to elective orchiectomy and the sperm will be obtained directly from the tail of the epidymidis and the vas deferens. The animals will be divided into 18 pools with the material of three animals each and there will be formed four experimental groups. In the control group (G0) semen will be freeze with the Botu-crio® diluents. In groups A (GA), B (GB) and C (GC), Botucrio diluents will be added with two antioxidants in different concentrations, 0,3, 0,6 e 0,9mM, respective. For each group, fresh semen samples will be first evaluated for motility, vigor and morphology to verify the minimum quality required for freezing. The same parameters will be re-evaluated after thawing and also for: Test Vital Supra, Hypoosmotic Test, Evaluation of Acrosomal Integrity and Lipid Peroxidation Reaction. The results will be evaluated using nonparametric analysis of variance and means will be compared by Tukey test (p <0.05).
