Effects of ovariectomy, estrogen and soy isoflavones in the acini and ducts of submandibular glands rats

Abstract

In postmenopausal women, there is a decrease in salivation, often associated with the burning mouth syndrome. Symptoms usually decline after estrogen therapy. There is a decrease of acini and granular ducts of submandibular glands in castrated rats, whereas, in rats with hormone replacement therapy and phototherapy, this effect is reversed. This study aims to elucidate mechanisms by which the changes occur in the acini and ducts in submandibular glands of rats castrated and given estrogen replacement therapy and/or soy isoflavones. This project is associated with another research, so the 75 rats have been operated on, treated, and euthanized. Of these, 60 were ovariectomized and 15 false-operated (Sham). Of ovariectomized, 15 received 17 b-estradiol, 15 received 15mg/kg/day extract 40% isoflavones, 15 received a combination of both orally, and 15 received water as a placebo. The sham-operated rats received water orally. The animals were killed three, five and eight weeks after ovariectomy. The submandibular and sublingual salivary glands were removed together, fixed, processed, and embedded in paraffin blocks. Blocks will be cut and stained with HE. Also will be performed immunohistochemistry streptavidin-biotin-peroxidase. It will be assessed the cell area histomorphometric and immunohistochemical expression of PCNA and estrogen receptor-b. The results will be statistically analyzed by ANOVA tests two factors and multiple comparison test of Tukey. (AU)
