Abstract

To create an artificial tissue in the laboratory is necessary an adequate support structure (scaffold), growth factors and cells. Currently, there is not an ideal bladder substitute and many research centers are trying to create a functional bladder. The bladder's lamina propria is composed by collagen, elastin, laminin, cells and bioactive factors secreted by these cells. The aim of this study is to evaluate the effect of lamina propria cells on the proliferation of urothelial cells and smooth muscle cells. Material and Methods: We will use twelve pigs, which will be undergone to partial cystectomy. We will perform the detrusor, urothelium and lamina propria micro dissection and the bladder's lamina propria will be decellularized and treated with mitomycin C. Lamina propria cells will be seeded on fibrin porous membrane to assess the paracrine effect. The evaluation of the growth and proliferation urothelium and smooth muscle cells in vitro will be carried out under three different conditions: urothelial cells and / or smooth muscle cells seeded on autologous decellularized bladder's lamina propria treated with Mitomycin C, urothelial cells and / or smooth muscle cells seeded in fibrin membrane previously seeded with lamina propria cells and urothelial cells and / or smooth muscle cells seeded in culture plates (controls).
