Abstract

The high incidence and mortality of hepatocellular carcinoma (HCC), as well as the limited treatment and poor prognosis of this disease make it important to adopt measures such as chemoprevention. Tributyrin (TB), a prodrug of butyric acid (BA) is a bioactive compound found in foods such as honey and milk. TB reduced the incidence and size of preneoplasic lesions and also induced apoptosis and histone acetylation when administered during the initiation and promotion or only during promotion phase of experimental hepatocarcinogenesis. However, its organoleptic characteristics and bioavailability limit the use of this substance as a chemopreventive agent. By an enzymatic reaction with specific 1,3 lipases, a mixture of TB and flax seed oil might increase the absorption and bioavailability of AB present in TB. Flaxseed oil is a natural source of ALA, and also has chemopreventive activity. The action of histone deacetylases inhibitors (HDAC), such as TB, has been related to changes in the methylation state of promoters. In this context, the objective of this project is to produce structured lipids by interesterification of TB with flax seed oil and to evaluate its chemopreventive activity in a model of hepatocarcinogenesis in rats. The expression of RASSF1A, NORE1A (RAASF5) and p21, as well as histone acetylation and alteration of DNA methylation, would elucidate the mechanisms and chemopreventive potential of the strutured lipids.
