Abstract

The aim of this project is to evaluate the specific phenomena of cell proliferation and cell death (apoptosis) in the liver after treatment with a mixture of five pesticides in NOEL (Mixture 1) or Effective Doses (LEL/LOEL/LOAEL) (Mixture 2). The pesticides (dieldrin, dicofol, endosulfan, dichlorvos, and permethrin), whose residues were detected in tomatoes by ANVISA in 2004, were added to the basal diet for treatment of male Lewis rats following two different experimental designs. In the first protocol (Design A), male Lewis rats were treated for eight weeks, according to the initiation (diethylnitrosamine-DEN)/ promotion hepatocarcinogenesis assay (Ito et al.,1988). In the second (Design B), the rats were treated for eight weeks, from six weeks of age, only with the feed containing the mixtures of pesticides at NOEL or LOEL doses (Mixtures 1 and 2 respectively). The proliferation index will be evaluated using immunohistochemical staining by anti-KI-67 (number of immunostained hepatocytes/ total hepatocytes analyzed x100) and apoptosis index by marking anti-cleaved Caspase 3 (number of immunostained cells/cm²).(AU)
