Abstract The aim of this study is test the hypothesis that dental pulp stem cells (DPSC) differentiate into vascular endothelial cells through regulation of Wnt/B-catenin ligands LRP6 or Frizzled6. DPSC will be transduced with EGFP-tagged lentiviral sh-RNA vectors (LRP6, Frizzled6 or empty vector) for experiments. Frizzled6 and LRP6 effects will be evaluated by GSK-3beta and Beta-catenin expression, and by endothelial marks expression in the differentiation of DPSCs into endothelial vascular cells. To validate this results, transduced DPSC will be seed in tooth slices/scaffolds and implanted subcutaneously in immunosupressed mice. Cell presence in blood vessels will be determined by immunohistochemestry and HE staining. | |
