Transgenerational epgenetic effect of high-sodium intake in Wistar rats

Abstract

To identify possible epigenetic alterations caused by the consumption of food with high levels of NaCl (salt), and if these alterations could affect germ cell lineage leading to next generation transmission, configuring transgenerational epigenetic alteration, Wistar female rats, on gestational phase, will be submitted to high-sodium concentration diet. The offspring will receive normosodic diet. Three generations will be accompanied (F1 to F3) to evaluate the heredity of possible alterations caused by this high-sodium consumption by F0. All generations will be monitored with blood and urinary concentrations of sodium, potassium and creatinin, albuminuria, hematocrit, aldosterone, plasmatic rennin activity and blood angiotensin conversion enzyme 1 and 2 activity (ACE1 and ACE2). Will be realized yet kidney and testicular histology besides expression and methylation of ECA-1 and HSD11 genes, that are related to both renal and reproductive factors. With this we intend to identify possible targets of epigenetic alterations caused by high-sodium intake, a very frequent condition in the majority of world populations, directing new procedures that could reverse this situation.
