Analysis of inflammasome in melanoma: correlation between tumoral cells and immune response mediated by dendritic cells

Abstract

It has been demonstrated that the progression of melanoma is closely related to an inflammatory microenvironment. In this sense, IL-1ß is an important pro-inflammatory mediator capable of coordinating the expression of other cytokines and chemokines, and mediate the recruitment of cells to the site of injury. In turn, the maturation of IL-1ß as well as its secretion is controlled by a cytoplasmic molecular complex known as inflammasome. The inflammasomes are expressed mainly in cells of innate immunity and dendritic cells (DCs). DCs have a major role in the driving the specific immune response that depends on the characteristics of DCs and the microenvironment of occurrence of antigen presentation. In this context, this project aims to investigate the impact of inflammasome on the immune response against melanoma through the analysis of its activation in monocyte-derived DC and in biopsies from melanoma patients. The genetic susceptibility will also be investigated by analyzing polymorphisms in genes of inflammasome in a cohort of melanoma patients. We expected that the results obtained from this project can provide information regarding the pathogenetic mechanisms involved in melanoma development and in the immune response against this type of cancer, as well as discover novel targets for future therapeutic approaches.
