Analysis of the effects of chronic fluoride intake in pathways that modulate angiogenesis and antioxidant system

Abstract

Excessive fluoride (F) intake over a long period of time could result in fluorosis, which can cause dental and skeletal manifestations. Metabolic, functional and structural damages caused by chronic fluorosis have been reported in many tissues. The kidneys are the most responsibility by fluoride excretion, around 60% of fluoride is absolved is excretion by urine in adults. Fluoride appears to influence oxidative stress and antioxidant system. Given this, the objective of this study will be to evaluate the effect of fluoride in antioxidant system in rat kidneys. Two groups of Wistar rats are used (n=24), were divided into two time periods (20 and 60 days), which will be divided into a control group (water), 15 ppm and 50 ppm fluoride experimental groups, which will be administered through drinking water. After experimental period, the animals will be euthanized and kidneys will be collected and divided to the following analysis: histomorphological analyzis, antioxidants analyzis (SOD, CAT, GPx, GSH, TBARS) and fluoride. Furthermore, enzymes are quantified VEGF, HIF-1±, EPO and GAPD-H in kidneys sample by Western Blotting. The results will analyzed according to appropriate statistical tests (p <0.05).
