Abstract

Diabetes mellitus during pregnancy is associated with higher levels of urinary incontinence and pelvic floor muscle dysfunction. This project will analyze the rectus abdominis muscle of pregnant rats with severe diabetes (blood glucose levels higher than 300 mg / dL) and moderate (blood glucose levels between 120 and 300 mg / dL). Wistar rats will be divided into six groups (n = 13 animals / group): non-diabetic and non-pregnant (control), non-diabetic pregnant, moderate diabetic non-pregnant, severe diabetic non-pregnant, moderate diabetic pregnant and severe diabetic pregnant. Moderate diabetes is induced on day 1 of birth by subcutaneous administration of streptozotocin at a dose of 100 mg / kg of body weight diluted in 0.1 mol / l citrate buffer (pH 4.5). The inclusion criterion will be blood glucose levels between 120 and 300 mg / dL. Severe diabetes is induced in adulthood (90 days of life) by intravenous administration of streptozotocin at a dose of 40 mg / kg body weight. The inclusion criterion will beblood glucose levels above 300mg/dL. At the end of the experiment, the rats will be anesthetized, euthanized and the rectus abdominis muscle will be removed, frozen in liquid nitrogen and stored at -80 ° C. The muscle will undergo in cryostat sections (6 mm thick) and will be submitted to cytochemical staining for muscle and components of the extracellular matrix, immunohistochemical labeling for slow and fast fibers and transmission electron microscopy for analysis of abdominal muscles. It will be analyzed the type and location of fast and slow muscle fibers of the rectus abdominis, the area occupied by connective tissue around the muscle fibers of the rectus abdominis and ultrastructure of striated muscle fibers of the rectus abdominal muscle and extracellular matrix. For statistical comparisons, will be considered the limit of significance p <0.05.
