Seminal quality and lipid peroxidation quantification of fresh and frozen-thawed cat semen using tris/yolk/glycerol or ACP 117®

Abstract

We aim to evaluate the quality of fresh and frozen-thawed semen in domestic cats using two different extenders (tris/yolk/glycerol and ACP 117®/ yolk/glycerol) and to quantify the lipid peroxidation of spermatic membranes before and after the thawed of semen. For that we will use three tom cats and one queen. The semen will be collected through artificial vagina. Right after the semen collection, the seminal evaluation will be performed, observing the macro and microscopic characteristics. For the macroscopic evaluation, we will measure the volume and observe the color of the semen. Microscopically we will evaluate the semen morphology, kinetic and concentration. One sample of fresh semen, contenting 5x106 spermatozoa will be used for lipid peroxidation quantification, using the TBARS method, described Buege e Aust (1978). After this evaluation the semen will be separated in two samples (A and B), the A sample will be extended using tris/yolk (20%)/glycerol(4%) and the B sample will be extended using ACP 117®/ yolk (20%)/glycerol(4%), both will be performed aiming to obtain the final concentration of 20x106 spermatozoa/mL. The semen will be placed in 0.25ml straw, and will be refrigerated at 5o C during 40 min, after this they will be transferred to a cooler with nitrogen and will be under nitrogen vapor, then this stews will be submerged in nitrogen and placed in canisters. The samples will be thawed in 38oC during 60 seconds and will be evaluated has described before. The data will be tabulated and submitted to ANOVA, and Tukey test for the media comparison, the results are significant different when P<0,05.(AU)
