Abstract

Cryo-Electron Microscopy (cryo-EM) is a powerful imaging tool to reveal the three-dimensional (3D) structure of samples at the molecular and cellular levels. Grunewald's group is pioneer to apply this technics to study macromolecular complexes in their native environment. This technology can be applied for a range of systems: eukaryotic cells, entire prokaryotic cells, organelles and bacteria cellulosomes. The aim of this project is to understand the underlying mechanistic properties of membrane remodeling based on the in vitro study of UL31 and UL34 proteins with membrane model using fluorescence microscopy and visualize in Cryo-electron tomography. This is an excellent model system for studying macromolecular interactions and its effects on cell membrane in the course of their function. Our goal is to integrate high-resolution structural information with imaging technics to achieve a spatial-temporal description of complex biological processes to reveal native structure-function relationships.
