Effect of Tyrosol on adhesion capacity of Candida cells to acrylic resin surface

Abstract

This study aims to evaluate the effect of different Tyrosol concentrations on adhesion capacity of Candida albicans and Candida glabrata to acrylic resin surface. First, surfaces of the acrylic resin specimens (10 x 10 x 3 mm) will be characterized by the determination of surface roughness and by the hydrophobicity degree. It will also be determined the hydrophobicity degree of yeast cell surfaces tested. After, the stock solution of Tyrosol will be diluted in inoculum of each yeast (107 cells/mL in artificial saliva) in order to obtain different Tyrosol concentrations. Each dilution (1.5 mL) will be added to the wells of 24-wells microtiter plates containing acrylic resin specimens. The plates will be incubated at 37°C for 2 hours with agitation of 120 rpm. Subsequently, the effect of Tyrosol on adhesion capacity of Candida species will be quantified by counting the number of culturable cells, total biomass (crystal violet staining assay) and metabolic activity of the cells. Adhered cells to acrylic surfaces after exposure to Tyrosol will be visualized by scanning electron microscopy. Controls devoid Tyrosol will be included in all analyzes, and chlorhexidine digluconate will be used as a positive control. All tests will be performed in triplicate on three different occasions. Data will be analyzed to employ the most appropriate statistical test. P < .05 will be considered statistically significant.
