Effect of TiF4 varnish on the progression of dentin erosion when applied in the presence or absence of the demineralised organic layer

Abstract

The aim of this in vitro study is to compare the effect of TiF4 and NaF varnish, with equivalent fluoride concentration, applied on bovine dentin samples pre-eroded in respect to the progression of wear, in the presence or absence of the demineralized organic layer (DOL). For that, 160 bovine dentin samples will be obtained from the cervical region of the root, polished and subjected to analysis of the baseline profile. The samples will be then eroded for 30 minutes with 0.1% citric acid (pH 2.5). Thereafter, the profile of eroded surface will be obtained. Half of the samples will be subjected to the removal of the DOL using collagenase for 5 days, while the other half will be immersed in water (control, maintenance of the DOL). The removal of DOL will be checked by profilometry (DOL profile). Samples with and without DOL will be randomly divided into 4 groups (n= 20 with and 20 without DOL/group): Group 1 - TiF4 varnish (2.45% F, pH 1); Group 2 - NaF varnish (2.45% F, pH 5); Group 3 - placebo varnish/without fluoride (pH 5); Group 4 - control (no treatment). The varnishes will be applied in thin layer on the tooth surface and removed after 6 hours. In the sequence, the samples will be submitted to a pH cycling for 5 days: 0.1% citric acid (4 x 90s/day) and the remineralization by immersion in artificial saliva between the erosive challenges. The final profile will be obtained in the presence and absence of DOL. The baseline, eroded, DOL and final profiles will be overlaid to calculate the degree of wear (µm). The data will be submitted to appropriate statistical tests (p <0.05, n=15). Five dentin samples per group will be analyzed by scanning electron microscopy.
