Emerging enteric viruses in wastewater from São Paulo city: Epidemiological surveillance

Abstract

Several infectious diseases can be transmitted through water contaminated with fecal material, including hepatitis and viral gastroenteritis. Among the enteric viruses, the most frequent target of research in the environment are norovirus (NoV), rotavirus species A (RV-A), human adenovirus (HAdV) and hepatitis A virus (HAV). Currently, the occurrence of other emerging viruses, such as polyomavirus (JCPyV) and hepatitis E virus (HEV) is also performed. The aim of this project is to establish a an environmental surveillance of these microorganisms in raw and treated sewage (including reclaimed water) from wastewater treatment plants (WWTPs) of the São Paulo city. Bacteriophage Bacteroides GB-124, potential indicator of the presence of these viruses in sewage, will also be analyzed by phage lyses technique. Secondary and tertiary treatment processes (filtration, ultra filtration and chlorination) will be evaluated for virus removal, allowing characterization of virological quality of effluents and the risks associated with reuse practices or disposal in surface water. Techniques of nucleic acid amplification and molecular characterization by nucleotide sequencing and phylogenetic analysis will be used for detection, quantification and viral genotyping. These results will provide relevant information about the epidemiology of enteric and emerging viruses (NoV GIV, and JCPyV HEV) in wastewater from São Paulo city Analytical protocols for viral detection will be tested and validated in order to provide useful diagnostic tools to assess emerging pathogens in the environment.
