Mutational evaluation of BRCA1 and BRCA2 genes in breast tumors in cats and correlation with clinical, histopathological and immunohistochemical aspects

Abstract

Breast cancer is a neoplasia with great importance in women and also has a high prevalence in small animals. These tumors are classified histologically and sub classified according to immunohistochemical and molecular markers, which are directly related to predictive and prognostic factors in humans. In cats, the majority of breast tumors do not express receptors for estrogen (ER) and progesterone (PG), as well has no overexpression of epidermal growth factor 2 (HER-2), being classified as triple-negative tumors. In women, breast tumors with this same phenotype are considered more aggressive, resistant to the therapies instituted and commonly have mutations in the BRCA1 and BRCA2 genes. Knowing that breast tumors in cats are mostly malignant and with a high degree of aggressiveness, this project aims to investigate the presence of mutations in the BRCA1 and BRCA2 genes, correlating this data with those clinical, histopathological and immunohistochemical. Will be included in the study 52 cats submitted to medical appointment and surgical procedure in the Surgery Service of Public Veterinary Hospital of São Paulo. Clinical and laboratory data will be obtained from medical records of each animal. Histological evaluation will be conducted, besides immunohistochemistry, DNA extraction from paraffin-embedded material, with subsequent Polymerase Chain Reaction (PCR) and sequencing of several exons of BRCA1 and BRCA2 feline genes. The searching of new immunohistochemical and molecular markers in feline breast cancers would provide relevant information for the clinical management of these patients, allowing a better understanding of its pathogenesis as well as studies in comparative oncology.
