Abstract

The project aims to obtain an E. coli strain expressing the Caulobacter Hfq protein fusioned to an FLAG epitope-tag, as well as a Caulobacter conditional hfq mutant strain. The methods used will be PCR amplification and cloning in suitable plasmid vectors, induction of the protein expression and purification by affinity chromatography in nickel column, besides bacterial transformation and other Microbiology and Molecular Biology procedures.
