Abstract

Chronic venous ulcer is a disease that intensively affects the quality of life of patients and society as a whole, due to the degree of disability and public spending on treatment - which is not done properly in most Health units, especially due to the lack of knowledge about its pathophysiology and therapeutic methods. Studies on the etiology of this disease point imbalance between synthesis and degradation of extracellular matrix, collagen abnormal metabolism, early senescence of fibroblasts and "sequestration" of growth factors. Therapies have been developed aiming especially at curatives which interact with the wound environment, promoting its healing. This is the case of oxidized regenerated cellulose (in its acronym, ORC)/collagen, whose function is to re-balance ECM synthesis and degradation by regulating the amount of metalloproteinase(MMPs) and their inhibitors (TIMPs) in the wound bed, allowing thus better functioning fibroblasts. This study aims to evaluate the in vitro behavior of fibroblasts derived from venous ulcer bed in the presence of ORC / collagen as to their morphology, viability and synthesis of collagen type I, collagen type III TGF-², MMP-1, MMP -3, TIMP-1 and TIMP-3. Those cells will be submitted to ORC solution /collagen and evaluated by electron microscopy, MTS assay, proliferation curve of fibroblasts and immunofluorescence tests.
