Abstract

Septic encephalopathy is among the most serious disease affecting the central nervous system (CNS). It happens in 71 percent of septic patients and symptoms include: Altered state of consciousness, going from delirious to coma, convulsion, long-term cognitive impairment and functional disability among survivors. Contributes markedly to septic encephalopathy full progression: Inflammation and oxidative stress. Gold nanoparticles (AuNPs) have tremendous potential in combating that disease once they have intrinsic antiinflammatory and antioxidant effect. Thus, we aim to study the effect of AuNPs treatment in an animal model of septic encephalopathy. In order to that, sepsis will be induced in mice using the cecum perforation and ligation model. Mice will be treated intravenously (IV) with AuNPs-cit or AuNPs-IgG (10 to the power of 12 particles/mL), and as AuNPs control treatment, anti-H3.3 antibody solution, used to prepare AuNPs-IgG, or with sodium aurothiomalate (Sigma-Aldrich), in an antiinflamatory dose, 0.7 ug/g body weight, or with saline (control group), injected once, 2 or 4 hours after sepsis induction. The following parameters will be measured 6 hours after sepsis induction: BBB permeability, cytokine and cyclooxygenase (COX) products in brain, leukocyte and platelets blood count, markers of coagulation and platelet activation and leukocyte and platelet count in pial vessels.
