Abstract

L-asparaginase enzyme (ASNase) is responsible for converting L-asparagine aminoacid to L-aspartic acid and ammonia. This enzyme has important applications especially in the pharmaceutical industry, where it is used as an antileukemic biopharmaceutical and in food industry, applied to mitigate the formation of acrylamide, a highly toxic compound formed in some processed foods at high temperatures. The search for new ASNase producing microorganisms is important for obtaining enzymes with properties that allow a better biological activity and/or fewer adverse effects, and lower production costs. The Rhodotorula glutinis yeast is a possible producer of ASNase enzyme periplasmic nature. Due to its nature, this project aims to establish techniques for the extraction of periplasmic ASNase without significant loss of its biological activity. Then, will be evaluated cell disruption testing of yeast in question. It will be evaluated the disrputing of Rhodotorula glutinis cells by different techniques, particularly those involving the use of ionic liquids for extraction of L-asparaginase enzyme present in their periplasmic space. The project has a great importance for the characterization of yeast ASNase glutinis Rhodotorula, targeting this enzyme applications in the food and pharmaceutical industries. (AU)
